The Scientific Imperative of NAD+ and Sirtuin Activation

It protects normal human dermal fibroblasts from UVA-induced cytotoxicity and senescence by decreasing matrix metalloproteinase-1 (MMP-1) expression [15]. Mechanistically, apigenin can suppress mTOR activity and boost autophagosome formation, activating autophagy by preventing PI3K pathway activity [35]. Furthermore, apigenin acts as a senomorphic agent, blocking the transition of acute stress-associated phenotypes to the senescence-associated secretory phenotype (SASP), thus mitigating age-related organ dysfunction [47].

The scientific understanding of NAD+ precursors and sirtuin activators is increasingly supported by preclinical and emerging clinical data, demonstrating a range of health and longevity benefits.

Nicotinamide Riboside (NR) and Nicotinamide Mononucleotide (NMN): In preclinical models, NR supplementation significantly improved mitochondrial function and rejuvenated muscle stem cells (MuSCs) in aged mice, preventing MuSC senescence and extending overall lifespan [14]. Recent research also indicates that NAD+ supplementation, specifically with NR, can reduce neuroinflammation, DNA damage, apoptosis, and cellular senescence in transgenic mouse models of Alzheimer’s disease by normalizing the cGAS-STING pathway. This further improved cognitive and synaptic functions [33]. Furthermore, topical and systemic applications of resveratrol have shown protective effects against UVA-induced photoaging in human skin fibroblasts and in the skin of male mice, indicating its potential in dermatological anti-aging strategies [41].

Apigenin: Beyond its anti-inflammatory and antioxidant properties, apigenin has demonstrated direct benefits for skin health and cellular senescence. In vitro studies showed apigenin restored viability and protected normal human dermal fibroblasts from UVA-induced senescence, while also decreasing the expression of MMP-1, a collagenase implicated in skin aging [15]. Clinical applications of apigenin-containing creams in human subjects resulted in increased dermal density and elasticity, along with a reduction in fine wrinkle length, improved skin evenness, and moisture content [15]. Apigenin’s senomorphic properties, which involve inhibiting the senescence-associated secretory phenotype (SASP), have been shown to improve physical function and alleviate frailty and cognitive impairment in mice, highlighting its potential in addressing age-related conditions and enhancing chemotherapeutic efficacy [47]. Its ability to modulate key molecular targets and signaling pathways also underscores its therapeutic potential against various cancers [28, 35].
